For most of us, managing our health means visiting a doctor. The more serious our concerns, the more specialized a medical expert we seek. Our bodies often feel like foreign and frightening lands, and we are happy to let someone with an MD serve as our tour guide. For most of us, our own DNA never makes it onto our personal reading list.

Biohackers are on a mission to change all that. These do-it-yourself biology hobbyists want to bring biotechnology out of institutional labs and into our homes. Following in the footsteps of revolutionaries like Steve Jobs and Steve Wozniak, who built the first Apple computer in Jobs’s garage, and Sergey Brin and Larry Page, who invented Google in a friend’s garage, biohackers are attempting bold feats of genetic engineering, drug development, and biotech research in makeshift home laboratories.

In Biopunk, journalist Marcus Wohlsen surveys the rising tide of the biohacker movement, which has been made possible by a convergence of better and cheaper technologies. For a few hundred dollars, anyone can send some spit to a sequencing company and receive a complete DNA scan, and then use free software to analyze the results. Custom-made DNA can be mail-ordered off websites, and affordable biotech gear is available on Craigslist and eBay.

New insight into factors that drive muscle-building stem cells

A report in the January issue of Cell Metabolism, a publication of Cell Press, provides new evidence explaining how stem cells known as satellite cells contribute to building muscles up in response to exercise. These findings could lead to treatments for reversing or improving the muscle loss that occurs in diseases such as cancer and AIDS as well as in the normal aging process, according to the researchers.

Cognitive ‘fog’ of normal aging linked to brain system disruption

Comparisons of the brains of young and old people have revealed that normal aging may cause cognitive decline due to deterioration of the connections among large-scale brain systems. The researchers linked the deterioration to a decrease in the integrity of the brain’s “white matter,” the tissue containing nerve cells that carry information. The researchers found that the disruption occurred even in the absence of pathology associated with Alzheimer’s disease (AD).

A longer-living, healthier mouse that could hold clues to human aging

A study by scientists at UCL (University College London) shows that mice lacking the insulin receptor substrate (IRS)-1 are more resistant to ageing than normal mice. The research adds to a growing body of work showing the importance of insulin signalling pathways as an ageing mechanism in mammals – and potentially humans.

Zinc may reduce pneumonia risk in nursing home elderly

When elderly nursing home residents contract pneumonia, it is a blow to their already fragile health. Simin Nikbin Meydani, DVM, PhD of the Jean Mayer USDA Human Nutrition Research Center on Aging at Tufts University and colleagues report that maintaining normal serum zinc concentration in the blood may help reduce the risk of pneumonia development in that population.

Scientists reveal how caloric restriction extends life by reducing toxic trash

Reduce, recycle and rebuild is as important to the most basic component of the human body, the cell, as it is to the environment. And a University of Florida study shows just how much the body benefits when it “goes green,” at least if you’re a rat: Cutting calories helps rodents live longer by boosting cells’ ability to recycle damaged parts so they can maintain efficient energy production.

“Caloric restriction is a way to extend life in animals. If you give them less food, the stress of this healthy habit actually makes them live longer,” said Christiaan Leeuwenburgh, chief of the division of biology of aging in UF’s Institute on Aging. Understanding how the process works at the cellular level in rodents could help scientists develop drugs that mimic the process in humans, Leeuwenburgh added.

Loneliness is bad for your health

Two University of Chicago psychologists, Louise Hawkley and John Cacioppo, have been trying to disentangle social isolation, loneliness, and the physical deterioration and diseases of aging, right down to the cellular level.

The researchers suspected that while the toll of loneliness may be mild and unremarkable in early life, it accumulates with time. To test this idea, the scientists studied a group of college-age individuals and continued an annual study of a group of people who joined when they were between 50 and 68 years old.

Their findings, reported in the August issue of Current Directions in Psychological Science, a journal of the Association for Psychological Science, are revealing. Consider stress, for example. The more years you live, the more stressful experiences you are going to have: new jobs, marriage and divorce, parenting, financial worries, illness. It’s inevitable.

A search for the molecular clues of longevity has taken Mayo Clinic researchers down another path that could explain why some people who consume excessive calories don’t gain weight. The study, which was done in laboratory mouse models, points to the absence of a gene called CD38. When absent, the gene prevented mice on high-fat diets from gaining weight, but when present, the mice became obese.

It’s not too late to change and lower cardiac risk later in life

Can adopting a healthier lifestyle later in life help — or is it too late? In a study published in the July 2007 issue of The American Journal of Medicine, researchers from the Medical University of South Carolina, Charleston found that people 45 to 64 years of age who added healthy lifestyle behaviors could substantially reduce their risk for cardiovascular disease (CVD) and reduce their death rate. Once these people achieved 4 healthy behaviors, eating at least 5 fruits and vegetables daily, exercising at least 2.5 hours per week, maintaining their Body Mass Index (BMI) between 18.5 and 30 kg/m, and not smoking, investigators saw a 35% reduction in CVD incidence and a 40% reduction in mortality compared to people with less healthy lifestyles.

Scientists find key to stem cell immortality

One of the medical marvels of stem cells is that they continue to divide and renew themselves when other cells would quit. But what is it that gives stem cells this kind of immortality. Researchers now report in the June 16, 2005 issue of the journal Nature that microRNAs — tiny snippets of genetic material that have now been linked to growth regulation in normal cells as well as cancer growth in abnormal cells — appear to shut off the “stop signals” or brakes that would normally tell cells to stop dividing.

Higher intake of fish and vitamin D levels linked to lower risk of age-related macular disease

Individuals who have higher dietary intake of foods with omega-3 fatty acids and higher fish consumption have a reduced risk of advanced age-related macular degeneration, while those with higher serum levels of vitamin D may have a reduced risk of the early stages of the disease, according to two reports in the May issue of Archives of Ophthalmology, one of the JAMA/Archives journals.

It’s never late to reduce calorie intake and slow aging

Much research has shown that reduced calorie intake can increase health and longevity. Professor Stephen Spindler (University of California) and his collaborators have discovered that reducing calorie intake later in life can still induce many of the health and longevity benefits of life-long calorie reduction. Importantly, this also includes anti-cancer effects. They are using this knowledge to establish a novel screening technique to find drugs which mimic this longevity effect.

How Eating Less Might Make You Live Longer

Caloric Restriction in non-obese people translates into less oxidative damage in muscle cells, according to a new study by scientists at Pennington Biomedical Research Center. As oxidative damage has been linked to aging, this could explain how limiting calorie intake without malnutrition extends life span.

Oxidative damage in newly synthesized DNA plays a role in Parkinson’s disease

Oxygen is the quintessential two-edged sword of molecular biology: essential for (animal) life, but at the same time a perennial source of damage to macromolecules. Reactive oxygen species (ROS), arising from both external sources and the intrinsic metabolic machinery of the cell itself, have been implicated in many aspects of cellular aging.

Of particular interest to human beings, especially those living in the rapidly aging post-industrial Western nations, is the relationship between oxidative damage and neurodegenerative illness. While most of the age-related neurodegenerative diseases are caused by accumulation of protein aggregates, it is becoming evident that ROS play an important role in exacerbating the underlying pathologies: e.g., DNA oxidation arises early in the pathogenesis of Alzheimer’s disease; and oxidative damage to a key anti-oxidant defense protein may generate a pernicious positive-feedback loop in the initiating events of Parkinson’s disease.

Whiff of Food Cancels Longevity from Caloric Restriction

Evidence began mounting as long as 70 years ago that restricting calories while consuming necessary amounts of sustenance could increase one’s life span. Since then, a group called the North Carolina-based Calorie Restriction Society has sprouted whose 1,800 members routinely down about half of the daily caloric intake recommended by the Food and Drug Administration (FDA) in the hope of living to the ripe old age of 120.

New research may prompt the organization to send out nose plugs with its next newsletter.A team of scientists at the Baylor College of Medicine in Houston, found that the average life span of fruit flies on restricted diets decreased when they were exposed to food odors.

Inflammation is culprit in many ailments

Chronic inflammation spurred by an immune system run amok appears to play a role in medical evils from arthritis to Alzheimer’s, diabetes to heart disease. There’s no grand proof of this “theory of everything.” But doctors say it’s compelling enough that we should act as if it were true — which means eating an “anti-inflammatory diet,” getting lots of physical activity, and losing the dangerous, internal belly fat that pumps out the chemicals that drive inflammation.

This is a year old article but well written to summarize the potency of inflammation as source of age-related damage.

Flavanols in cocoa may offer benefits to the brain

A special cocoa made to retain naturally occurring compounds called flavanols may have the potential to help maintain healthy brain function and chart the course for future research that could lead to new solutions for preventing cognitive decline and dementia, according to a panel of scientists who presented new data at the annual meeting of the American Association for the Advancement of Science (AAAS).

Several studies suggest that consumption of a special cocoa made to be rich in flavanols, a naturally occurring nutrient abundant in fresh cocoa, may improve blood vessel function. Now, scientists believe the potential blood flow benefits associated with consumption of this flavanol-rich cocoa may extend to the brain — which could have important implications for learning and memory.

Does inhibiting a component of niacin point the way to anti-aging drugs?

In recent years, scientists have discovered that a family of enzymes called sirtuins can dramatically extend life in organisms as diverse as yeast, worms, and flies. They may also be able to control age-associated metabolic disorders, including obesity and type II diabetes.

Naturally occurring substances have been shown to activate sirtuins, including a constituent of red wine called resveratrol – although an individual would need to drink about two cases of wine a day to derive a clinically effective dose of resveratrol. Still, the findings have energized a number of scientific groups and biotechnology companies, all of which are now eagerly searching for drug candidates able to boost sirtuin activity. The public-health benefits of such an “anti-aging” drug would be substantial – as would the economic returns.

Now, a new study from scientists at The Wistar Institute points to another strategy for activating sirtuins to unleash their anti-aging powers.

Surveyors of anti-aging elixirs tout human growth hormone as a remedy for all things sagging-from skin to libidos – and claim it can even prevent or reverse aging. But researchers at the Stanford University School of Medicine say there’s no evidence to suggest that this purported fountain of youth has any more effect than a trickle of tap water when it comes to fending off Father Time.

Scientists find new genetic clue to cause of Alzheimer’s disease

Variations in a gene known as SORL1 may be a factor in the development of late onset Alzheimer’s disease, an international team of researchers has discovered. The genetic clue, which could lead to a better understanding of one cause of Alzheimer’s, is reported in Nature Genetics online, Jan. 14, 2007, and was supported in part by the National Institutes of Health (NIH).

The researchers suggest that faulty versions of the SORL1 gene contribute to formation of amyloid plaques, a hallmark sign of Alzheimer’s in the brains of people with the disease. They identified 29 variants that mark relatively short segments of DNA where disease-causing changes could lie. The study did not, however, identify specific genetic changes that result in Alzheimer’s.

Buildup of damaged DNA in cells drives aging

The accumulation of genetic damage in our cells is a major contributor to how we age, according to a study being published today in the journal Nature by an international group of researchers. The study found that mice completely lacking a critical gene for repairing damaged DNA grow old rapidly and have physical, genetic and hormonal profiles very similar to mice that grow old naturally. Furthermore, the premature aging symptoms of the mice led to the discovery of a new type of human progeria, a rare inherited disease in which affected individuals age rapidly and die prematurely.

Longevity gene keeps mind sharp

A gene variation that helps people live to a ripe old age also appears to preserve memory and thinking power, US work suggests. The “longevity” gene alters the size of fatty cholesterol particles in the blood, making them bigger than normal.

This stops them causing the fatty build up in blood vessels that is linked with brain impairment, and deadly strokes and heart attacks, Neurology reports.

Living to be 100 programmed at birth?

A study from the University of Chicago has been getting a lot of press attention lately. They found that your chances of living to 100 may depend on how young your mother was when she gave birth to you with chances doubled if mom was under 25. The reports do not give the explanation why.

Dr. Steven Palter, a specialist in Obstetrics and Gynecology and Reproductive Endocrinology and Infertility, has written in his blog docinthemachine about an interesting theory of reproductive aging and human longevity. The findings are based on data presented in recent American Society for Reproductive Medicine meeting by Dr. Keefe of U. Florida on the telemore and its role in reproductive aging.

Resveratrol prevents obesity and drastically increases physical endurance.

Researchers at the Institute of Genetics and Molecular and Cellular Biology in Illkirch, France have found that resveratrol boosts the exercise capacity of muscles in mice and protects against diet-induced insulin resistance and obesity. Link identified between age, cardiovascular disease

Researchers in the Linus Pauling Institute at Oregon State University have discovered a fundamental mechanism that causes aging blood vessels to lose their elasticity – a literal “hardening of the arteries” that is often a prelude to high blood pressure and cardiovascular disease.

An understanding of this mechanism, scientists say, provides an important new target for both drugs and dietary changes that might help prevent or treat atherosclerosis and heart disease. This is a leading cause of death around the world that, in some form, affects about 80 percent of older Americans.

Scientists have identified a misfolded, or incorrectly formed, protein common to two devastating neurological diseases, frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS, also known as Lou Gehrig’s disease), according to a report in the Oct. 6, 2006, issue of Science. The findings suggest that certain forms of FTD, ALS and possibly other neurological diseases might share a common pathological process.

A new mechanism of aging discovered

We all age, it’s a fact of life, like death and taxes, and there’s nothing we can do about it. But, how is it possible that of two middle-aged mice, one is already grey, balding and frail? Some mice that age three times faster than normal are revealing to scientists why we grow old.

Researchers have discovered that genetic mutations in the powerhouses of our cells — mitochondria — appear to trigger cells to die and speed up the aging process. Inducing these kinds of mitochondrial mutations leads to premature aging in mice, which live only about half as long as normal mice.

Do Dieting Monkeys Live Healthier and Longer Lives?

An ongoing study at the University of Wisconsin-Madison in which rhesus monkeys are being fed an extremely calorie-restricted diet gives preliminary evidence that the regime prevents age-related diseases. For decades, scientists have known that a diet of about 30-percent fewer calories than normal extends the lifespan of mice by 10 to 20 percent, reduces their incidence of cancer, and prevents the deterioration of learning and memory in the rodents. And similar effects have been shown in lower organisms from yeast to fruit flies. But such life extension has not been proven yet in primates. Preliminary evidence from one of the largest studies of calorie-restricted diet in primates shows health benefits.

Studies find general mechanism of cellular aging

Three separate studies confirm a gene that suppresses tumor cell growth also plays a key role in aging. The researchers found increasing concentration, or expression, of the gene p16INK4a in older cells; these aging cells worked poorly compared to young cells and remembered their “age” even when transferred from old mice to young mice. The cells of mice bred without the gene showed less sluggishness as the animals aged and continued to function in a manner more similar to cells from younger mice.

Family Of Life-extending Genes Discovered

Mice, rats, worms, flies, and yeast all live longer on a low-calorie diet, which also seems to protect mammals against cancer and other aging-related diseases. A gene called SIR2 is thought to control this process. Now, researchers at Harvard Medical School and UC Davis have discovered four cousins of the SIR2 gene that also extend lifespan, suggesting that the whole family of SIR2 genes is involved in controlling lifespan. The research indicates potential targets for developing drugs to lengthen life and prevent or treat aging-related diseases. The findings are reported July 28 in the advance online edition of Science. This discovery comes on the heels of the Harvard group’s discovery of a molecule in red wine that extends the lifespan of every organism so far tested.

Researchers Find New Clues To Biochemistry Of ‘Anti-Aging

University of Wisconsin-Madison researchers have found that sirtuins, a family of enzymes linked to a longer life span and healthier aging in humans, may orchestrate the activity of other enzymes involved in metabolic processes in the body.
Published in the Proceedings of the National Academy of Sciences, the study is the first to show that sirtuins directly control specific metabolic enzymes – called AceCSs – in mammalian cells.

The finding, which shines a spotlight on enzymes only recently thought to play a role in the biochemistry of “anti-aging,” has attracted the interest of biotechnology companies seeking to make drugs that delay the aging process and age-related diseases. The drugs could target the metabolic enzymes to produce health benefits.

Scientists find rate of aging is at least in part genetically determined

We can dye gray hair, lift sagging skin or boost lost hearing, but no visit to the day spa would be able to hide a newly discovered genetic marker for the toll that time takes on our cells. “We’ve found something that is at the core of aging,” said Stuart Kim, PhD, professor of developmental biology and of genetics at the Stanford University School of Medicine.

In a study to be published in the July 21 issue of Public Library of Science-Genetics, Kim and colleagues report finding a group of genes that are consistently less active in older animals across a variety of species. The activity of these genes proved to be a consistent indicator of how far a cell had progressed toward its eventual demise.

Researchers discover new cell properties in accelerated aging process

Carnegie Mellon University researchers Kris Noel Dahl and Mohammad F. Islam have made a new breakthrough for children suffering from an extremely rare disease that accelerates the aging process by about seven times the normal rate. Researchers found that children suffering from Hutchinson-Gilford Progeria Syndrome (HGPS) have an excessively stiff shell of proteins.

Anti-Aging Molecule Discovered

A team of South Korean scientists on Sunday claimed to have created a “cellular fountain of youth,’’ or a small molecule, which enables human cells to avoid aging and dying.

The team, headed by Prof. Kim Tae-kook at the Korea Advanced Institute of Science and Technology, argued the newly-synthesized molecule, named CGK733, can even make cells younger.

"A common molecular denominator in aging and many age-related diseases is oxidative stress," says the study's lead author Azad Bonni, MD, PhD, HMS associate professor of pathology. The skin of a bitten apple will brown because of its exposure to air, and in some ways that is a good metaphor for the damage that oxidative stress is causing to neurons and other types of cells over time.

How the oxidative-stress signals trigger these profound effects in cells has remained unclear. But Bonni and his research team, have now defined how a molecular chain-of-events links oxidative-stress signals to cell death in brain neurons.

Mice that lives 20 percent longer

The canny world of advertising has caught on to the free radical theory of aging, marketing a whole array of antioxidants for preventing anything from wrinkles to dry hair to reducing the risk of heart disease — promising to help slow the hands of time.

Nevertheless, numerous studies of people taking antioxidant pills have failed to show a benefit, and the supplements may even be harmful. A study earlier this year hinted that high doses of the antioxidant vitamin E may raise the risk of heart disease, while earlier research has found that beta carotene, another popular antioxidant, puts smokers at higher risk of lung cancer. But that doesn’t mean the free radical theory of aging is wrong, “We think that it is fundamental to the understanding and the implications of aging,” says University of Washington pathologist, Peter Rabinovitch.

Working with genetically engineered mice — to produce a natural antioxidant enzyme called catalase — Rabinovitch’s group found that, on average, the mice live longer. But don’t go running to the medicine cabinet for your bottle of Vitamin C or other antioxidant supplement, only naturally-occurring antioxidants seem to offer a dip in the fountain of youth, and so far, only in mice.

Resveratrol prolongs lifespan in a short-lived vertebrate

By studying a particularly short-lived fish species, researchers have been able to show that a natural compound, Resveratrol, previously shown to extend lifespan in non-vertebrate organisms can also do so in at least one vertebrate species. The findings support the potential utility of the compound in human aging research.

Scientists have determined the detailed structure of an essential piece of the telomerase enzyme, an important contributor to the vast majority of human cancers. Understanding the physical shape of the protein has led to a better understanding of how it acts to immortalize cells – and should help scientists design broadly effective cancer drugs.

Researchers provide fresh evidence for aging cells causing aging body

Brown University biologists have uncovered intriguing evidence to support the theory that old cells help make old bodies. In a study of baboons, scientists showed that as these animals age, the number of aging cells in their skin significantly increases.

Over time, cells lose their ability to divide, a state known as replicative senescence. The new research, published in an advanced online edition of Science, is the first to quantify the presence of replicatively senescent cells in any species.
